Thiazolidinediones and fractures: evidence from translating research into action for diabetes

There can be no denying that thiazolidinediones (TZDs), in particular rosiglitazone, have received considerable bad press in recent years. One area of concern has been the association between TZDs and fractures, particularly fractures of small bones in post-menopausal women. There is a plausible mechanism to underlie this association in that activation of the peroxisome proliferator-activated receptor-γ can reduce bone formation in vitro and in vivo.
