Targeting the multiple myeloma hypoxic niche with TH-302, a hypoxia-activated prodrug.

Editor’s note: In solid tumors, hypoxia is associated with increased angiogenesis, metastatic potential, and a poorer outcome for patients. However, the role of hypoxia in the pathogenesis of hematological malignancies is unclear and indeed there is good evidence that hypoxia is crucial for normal hematopoiesis. In myeloma, it is known that the bone marrow (BM) environment can influence both the growth of the malignant clone and drug resistance.
