Editor’s note: The authors have previously investigated chronic myelomonocytic leukemia (CMML) and found chromosome region 11q23.3 to be an area of recurrent somatic uniparental disomy (UPD; CancerRes 2008;68:10349–57). The E3 ubiquitin ligase c-Cbl, which is involved in degradation of some of the key kinases of signal transduction, maps to this area. Mutations in c-Cbl were found frequently in CMML and secondary AML. Given some of the morphological and biological similarities between JMML and CMML in addition to the finding of UPD11q23.3 in JMML, the authors sought Cbl family mutations in JMML.
